EffectsoftheOralAdministrationofViableandHeat- KilledStreptococcusbovisHC5CellstoPre-Sensitized BALB/cMice AlineD.Paiva1,KennerM.Fernandes2,RobertoS.Dias2,Al?′pioS.Rocha2,LeandroL.deOliveira2, Clo′visA.Neves2,Se′rgioO.dePaula2,Hila′rioC.Mantovani1* 1DepartamentodeMicrobiologia,UniversidadeFederaldeVic?osa,Vic?osa,MinasGerais,Brazil,2DepartamentodeBiologiaGeral,UniversidadeFederaldeVic?osa,Vic?osa, MinasGerais,Brazil Abstract Antimicrobial peptides have been suggested as an alternative to classical antibiotics in livestock production and bacteriocin-producingbacteriacouldbeaddedtoanimalfeedstodeliverbacteriocinsinthegastrointestinal(GI)tractof ruminantandmonogastricanimals.Inthisstudy,viable(V)andheat-killed(HK)StreptococcusbovisHC5cellswereorally administered to pre-sensitized mice in order to assess the effects of a bacteriocin-producing bacteria on histological parametersandtheimmuneresponseoftheGItractofmonogastricanimals.TheadministrationofVandHKS.bovisHC5 cellsduring58daystoBALB/cmicedidnotaffectweightgain,butanincreaseingutpermeabilitywasdetectedinanimals receivingtheHKcells.ViableandheatkilledcellscausedsimilarmorphologicalalterationsintheGItractoftheanimals,but themostprominent effectsweredetected inthesmall intestine.The oraladministration of S.bovis HC5 also influenced cytokineproductioninthesmallintestine,andtheimmune-mediatedactivitydifferedbetweenVandHKcells.Therelative expressionofIL-12andINF-cwassignificantlyhigherinthesmallintestineofmicetreatedwithVcells,whileanincreasein IL-5,IL-13andTNF-aexpressionwasonlydetectedinmicetreatedwithHKcells.Consideringthatevenunderaconditionof severechallenge(pre-sensitizationfollowedbydailyexposuretothesamebacterialimmunogen)thegeneralhealthofthe animals was maintained, it appears that oral administration of S. bovis HC5 cells could be a useful route to deliver bacteriocinintheGItractoflivestockanimals.
