ErythrocyteandPorcineIntestinalGlycosphingolipids RecognizedbyF4FimbriaeofEnterotoxigenic Escherichiacoli AnneliesCoddens1,ErikValis2,JohnBenktander2,JonasAngstro? ¨m2,MichaelE.Breimer3,EricCox1, SusannTeneberg2* 1Laboratory of Veterinary Immunology, Faculty of Veterinary Medicine, Ghent University, Merelbeke, Belgium, 2Institute of Biomedicine, Department of Medical BiochemistryandCellBiology,UniversityofGothenburg,Go¨teborg,Sweden,3DepartmentofSurgery,SahlgrenskaUniversityHospital,Go¨teborg,Sweden Abstract EnterotoxigenicF4-fimbriatedEscherichiacoliisassociatedwithdiarrhealdiseaseinneonatalandpostweaningpigs.TheF4 fimbriae mediate attachment of the bacteria to the pig intestinal epithelium, enabling an efficient delivery of diarrhea- inducingenterotoxinstothetargetepithelialcells.TherearethreevariantsofF4fimbriaedesignatedF4ab,F4acandF4ad, respectively,havingdifferentantigenicandadhesiveproperties.Inthepresentstudy,thebindingofisolatedF4ab,F4acand F4adfimbriae,andF4ab/ac/ad-fimbriatedE.coli,toglycosphingolipidsfromerythrocytesandfromporcinesmallintestinal epithelium was examined, in order to get a comprehensive view of the F4-binding glycosphingolipids involved in F4- mediatedhemagglutinationandadhesiontotheepithelialcellsofporcineintestine.SpecificinteractionsbetweentheF4ab, F4acandF4adfimbriaeandbothacidandnon-acidglycosphingolipidswereobtained,andafterisolationofbinding-active glycosphingolipids and characterization by mass spectrometry and proton NMR, distinct carbohydrate binding patterns were defined for each fimbrial subtype. Two novel glycosphingolipids were isolated from chicken erythrocytes, and characterized as GalNAca3GalNAc?3Gal?4Glc?1Cer and GalNAca3GalNAc?3Gal?4GlcNAc?3Gal?4Glc?1Cer. These two compounds,andlactosylceramide(Gal?4Glc?1Cer)withphytosphingosineandhydroxyfattyacid,wererecognizedbyall three variants of F4 fimbriae. No binding of the F4ad fimbriae or F4ad-fimbriated E. coli to the porcine intestinal glycosphingolipidsoccurred.Ho
