FlavodiironProteinsinOxygenicPhotosynthetic Organisms:PhotoprotectionofPhotosystemIIbyFlv2 andFlv4inSynechocystissp.PCC6803 PengpengZhang,YagutAllahverdiyeva,MarionEisenhut,Eva-MariAro* DepartmentofBiology,PlantPhysiologyandMolecularBiology,UniversityofTurku,Turku,Finland Abstract Background:Flavodiironproteins(FDPs)compriseagroupofmodularenzymesthatfunctioninoxygenandnitricoxide detoxificationinBacteriaandArchaea.TheFDPsincyanobacteriahaveanextradomainascomparedtomajorprokaryotic enzymes.ThephysiologicalroleofcyanobacteriaFDPsismostlyunknown.Ofthefourputativeflavodiironproteins(Flv1–4) inthecyanobacteriumSynechocystissp.PCC6803,aphysiologicalfunctioninMehlerreactionhasbeensuggestedforFlv1 andFlv3. PrincipalFindings:WedemonstrateanovelandcrucialfunctionforFlv2andFlv4inphotoprotectionofphotosystemII (PSII) in Synechocystis. It is shown that the expression of Flv2 and Flv4 is high under air level of CO2 and negligible at elevated CO2. Moreover,therate of accumulationof flv2 andflv4 transcripts upon shift of cells fromhigh to lowCO2 is strongly dependent on light intensity. Characterization of FDP inactivation mutants of Synechocystis revealed a specific declineinPSIIcentersandimpairedtranslationoftheD1proteininDflv2andDflv4whengrownatairlevelCO2whereasat highCO2theFlvsweredispensable.Dflv2andDflv4werealsomoresusceptibletohighlightinducedinhibitionofPSIIthan WTorDflv1andDflv3. Significance: Analysis of published sequences revealed the presence of cyanobacteria-like FDPs also in some oxygenic photosyntheticeukaryoteslikegreenalgae,mossesandlycophytes.OurdataprovideevidencethatFlv2andFlv4havean importantroleinphotoprotectionofwater-splittingPSIIagainstoxidativestresswhenthecellsareacclimatedtoairlevel CO2.ItisconceivablethatthefunctionofFDPshaschangedduringevolutionfromprotectionagainstoxygeninanaerobic microbes to protection against reactive oxygen species thus making the sustainable function of oxygen evolving PSII possible.HigherplantslackFDPsanddistinctlydifferent
