Abstract
Umamitaste(correspondstosavoryinEnglish)iselicitedbyL-glutamate,typicallyasitsNasalt(monosodiumglutamate:
MSG),andisoneoffivebasictastequalitiesthatplaysakeyroleinintakeofaminoacids.Aparticularpropertyofumamiis
thesynergisticpotentiationofglutamatebypurinenucleotidemonophosphates(IMP,GMP).AheterodimerofaGprotein
coupled receptor, TAS1R1 and TAS1R3, is proposed to function as its receptor. However, little is known about genetic
variationofTAS1R1andTAS1R3anditspotentiallinkswithindividualdifferencesinumamisensitivity.Hereweinvestigated
theassociationbetweenrecognitionthresholdsforumamisubstancesandgeneticvariationsinhumanTAS1R1andTAS1R3,
andthefunctionsofTAS1R1/TAS1R3variantsusingaheterologousexpressionsystem.Ourstudydemonstratedthatthe
TAS1R1-372Tcreatesamoresensitiveumamireceptorthan-372A,whileTAS1R3-757Ccreatesalesssensitiveonethan-
757RforMSGandMSGplusIMP,andshowedastrongcorrelationbetweentherecognitionthresholdsandinvitrodose-
responserelationships.TheseresultsinhumanstudiessupportthepropositionsthataTAS1R1/TAS1R3heterodimeractsas
anumamireceptor,andthatgeneticvariationinthisheterodimerdirectlyaffectsumamitastesensitivity.
