MolecularCharacterizationofAntimicrobialPeptide GenesoftheCarpenterAntCamponotusfloridanus CarolinRatzka1*,FrankFo¨rster2,ChunguangLiang2,MariaKupper1,ThomasDandekar2, HeikeFeldhaar3,RoyGross1 1Department of Microbiology, Biocentre, University of Wu¨rzburg, Wu¨rzburg, Germany, 2Department of Bioinformatics, Biocentre, University of Wu¨rzburg, Wu¨rzburg, Germany,3DepartmentofAnimalEcologyI,UniversityofBayreuth,Bayreuth,Germany Abstract The production of antimicrobial peptides (AMPs) is a major defense mechanism against pathogen infestation and of particularimportanceforinsectsrelyingexclusivelyonaninnateimmunesystem.Here,wereportonthecharacterizationof threeAMPsfromthecarpenterantCamponotusfloridanus.Duetosequencesimilaritiesandaminoacidcompositionthese peptides can be classified into the cysteine-rich (e.g. defensin) and glycine-rich (e.g. hymenoptaecin) AMP groups, respectively.ThegeneandcDNAsequencesoftheseAMPswereestablishedandtheirexpressionwasshowntobeinduced bymicrobialchallenge.Wecharacterizedtwodifferentdefensingenes.Thedefensin-2genehasasingleintron,whereasthe defensin-1genehastwointrons.ThededucedaminoacidsequenceoftheC.floridanusdefensinsisverysimilartoother knownantdefensinswiththeexceptionofashortC-terminalextensionofdefensin-1.Thehymenoptaecingenehasasingle intronandaverypeculiardomainstructure.Thecorrespondingprecursorproteinconsistsofasignal-andapro-sequence followedbyahymenoptaecin-likedomainandsixdirectlyrepeatedhymenoptaecindomains.Eachofthehymenoptaecin domainsisflankedbyanEAEP-spacersequenceandaRR-siteknowntobeaproteolyticprocessingsite.Thus,proteolytic processingofthemultipeptideprecursormaygenerateseveralmatureAMPsleadingtoanamplificationoftheimmune response. Bioinformatical analyses revealed the presence of hymenoptaecin genes with similar multipeptide precursor structure in genomes of other ant species suggesting an evolutionary conserved important role of this gene in ant immunity.
