Despite numerous past demonstrations oftheefficiency of this technique, fewstudies haveattempted toemploy DNA barcodingmethodologiesonalargegeographicscale,particularlywithintropicalregions.Inthisstudywesurveycurrent andpotential speciesdiversity usingDNAbarcodeswithacollection ofmorethan9000individuals from163speciesof Neotropical bats (order Chiroptera). This represents one of the largest surveys to employ this strategy on any animal groupandiscertainly thelargesttodateforlandvertebrates. Ouranalysis documents theutilityofthistoolovergreat geographic distances and across extraordinarily diverse habitats. Among the 163 included species 98.8% possessed distinct setsofCOIhaplotypes making themeasily recognizable atthis locus. Wedetected onlyasingle case ofshared haplotypes. Intraspecific diversity intheregionwashighamongcurrently recognized species (meanof1.38%, range0– 11.79%)withrespecttobirds,thoughcomparabletootherbatassemblages.In44of163cases,well-supported, distinct intraspecific lineages were identified which may suggest the presence of cryptic species though mean and maximum intraspecific divergence werenotgoodpredictors oftheirpresence. Inallcases,intraspecific lineagesrequire additional investigation using complementary molecular techniques and additional characters such as morphology and acoustic data.OuranalysisprovidesstrongsupportforthecontinuedassemblyofDNAbarcodinglibrarie
