OralBiofilmArchitectureonNaturalTeeth VincentZijnge1,2,M.BarbaraM.vanLeeuwen3,JohnE.Degener2,FrankAbbas1,ThomasThurnheer4, RudolfGmu¨r4,HermieJ.M.Harmsen2* 1Center for Dentistry and Oral Hygiene, University of Groningen, Groningen, The Netherlands, 2Department of Medical Microbiology, University of Groningen, Groningen, The Netherlands, 3Department of Biomedical Engineering, University of Groningen, Groningen, The Netherlands, 4Institute of Oral Biology, University of Zu¨rich,Zu¨rich,Switzerland Abstract Periodontitisandcariesareinfectiousdiseasesoftheoralcavityinwhichoralbiofilmsplayacausativerole.Moreover,oral biofilms are widely studied as model systems for bacterial adhesion, biofilm development, and biofilm resistance to antibiotics,duetotheirwidespreadpresenceandaccessibility.Despitedescriptionsofinitialplaqueformationonthetooth surface,studiesonmature plaqueandplaquestructurebelowthegumarelimitedtolandmarkstudiesfromthe1970s, withoutappreciatingthebreadthofmicrobialdiversityintheplaque.Weusedfluorescentinsituhybridizationtolocalizein vivothemostabundantspeciesfromdifferentphylaandspeciesassociatedwithperiodontitisonsevenembeddedteeth obtainedfromfourdifferentsubjects.ThedatashowedconvincinglythedominanceofActinomycessp.,Tannerellaforsythia, Fusobacteriumnucleatum,Spirochaetes,andSynergistetesinsubgingivalplaque.Thelatterprovedtobenewwithapossibly importantroleinhost-pathogeninteractionduetoitslocalizationincloseproximitytoimmunecells.Thepresentstudy identifiedforthefirsttimeinvivothatLactobacillussp.arethecentralcellsofbacterialaggregatesinsubgingivalplaque, and that Streptococcus sp. and the yeast Candida albicans form corncob structures in supragingival plaque. Finally, periodontalpathogenscolonizealreadyformedbiofilmsandformmicrocoloniestherein.Theseinvivoobservationsonoral biofilmsprovideaclearvisiononbiofilmarchitectureandthespatialdistributionofpredominantspecies.
