WhilstitisrecognizedthatlevelsofbioactiveBRsarelikelyadjustedbycontrollingtherelativeratesofbiosynthesis andbycatabolism,fewfactors,whichparticipateintheseregulatoryevents,haveasyetbeenidentified.Previously wehaveshownthattheUDP-glycosyltransferaseUGT73C5ofArabidopsisthalianacatalyzes23-O-glucosylationof BRsandthatglucosylationrendersBRsinactive.ThisstudyidentifiestheclosesthomologueofUGT73C5,UGT73C6, asanenzymethatisalsoabletoglucosylateBRsinplanta. Results:Inacandidategeneapproach,inwhichhomologuesofUGT73C5werescreenedfortheirpotentialto induceBRdeficiencywhenover-expressedinplants,UGT73C6wasidentifiedasanenzymethatcanglucosylatethe BRsCSandBLattheir23-O-positionsinplanta.GUSreporteranalysisindicatesthatUGT73C6showsover-lapping,but alsodistinctexpressionpatternswithUGT73C5andYFPreporterdatasuggeststhatatthecellularlevel,bothUGTs localizetothecytoplasmandtothenucleus.Aliquidchromatographyhigh-resolutionmassspectrometrymethod forBRmetaboliteanalysiswasdevelopedandappliedtodeterminethekineticsofformationandthecatabolicfate ofBR-23-O-glucosidesinwildtypeandUGT73C5andUGT73C6over-expressionlines.Thisapproachidentifiednovel BRcatabolites,whichareconsideredtobeBR-malonylglucosides,andprovidedfirstevidenceindicatingthat glucosylationprotectsBRsfromcellularremoval.Thephysiologicalsigni
