The 2174G.C and 2572G.C polymorphisms in the IL-6 gene have previously been shown to modulate IL-6 levels. But the association between the 2174G.C and 2572G.C polymorphisms and the risk of CHD is still unclear. A meta-analysis of all eligible studies was carriedouttoclarifytheroleofIL-6genepolymorphismsinCHD. MethodsandResults:PubMed,EMBASE,Vip,CNKIandCBM-discweresearchedforeligiblearticlesinEnglishandChinese thatwerepublishedbeforeOctober2010.27studiesinvolving11580patientswithCHDand17103controlswereincluded. Ameta-analysiswasperformedfortheincludedarticlesusingtheRevMan5.0andStata10.0softwares.Overall,the2174C allelewasnotsignificantlyassociatedwithCHDrisk(ORs=1.04,95%CI=0.98to1.10)whencomparedwiththe2174Gallele intheadditivemodel,andmeta-analysisunderothergeneticmodels(dominant,recessive,CCversusGG,andGCversusGG) alsodidnotrevealanysignificantassociation.Onthecontrary,the2572Callelewasassociatedwithadecreasedriskof CHDwhencomparedwiththe2572Gallele(ORs=0.79,95%CI=0.68to0.93).Furthermore,analysesundertherecessive model(ORs=0.69,95%=0.59to0.80)andtheallelecontrastmodel(genotypeofCCversusGG,ORs=0.49,95%=0.35to 0.70)yieldedsimilarresults.However,statisticalsignificancewasnotfoundwhenthemeta-analysiswasrestrictedtostudies focusingonEuropeanpopulations,studieswithlargesamplesize,andcohortstudiesbyusingsubgroupanalysis. Conclusions: The 2174G.C polymorphism in the IL-6 gene is not significantly associated with increased risks of CHD. However, The 2572G.C polymorphism may contribute to CHD development.
