PredictionoftheMechanismofActionofFusaricidinon Bacillussubtilis Wen-BangYu,Chun-YunYin,YingZhou*,Bang-CeYe* LabofBiosystemsandMicroanalysis,StateKeyLaboratoryofBioreactorEngineering,EastChinaUniversityofScienceandTechnology,Shanghai,China Abstract Long-termuseofantibioticshasengenderedalargenumberofresistantpathogens,whichposeaseriousthreattohuman health.Here,weinvestigatedthemechanismoffusaricidinantibacterialactivitytowardBacillussubtilisandcharacterized the pathways responsible for drug resistance. We found that sw, an extracytoplasmic function sigma factor, plays an importantroleintheresistancetofusaricidinsduringtheinitial5minutesofdrugaddition.Approximately18geneswere inducedmorethan3-fold,ofwhich66.7%areknowntoberegulatedbysw.Overthefollowing3h,fusaricidinsinduced 194genesmorethanthree-fold,andmostwereassociatedwithclassesofantibiotic-responsivestimulons.Moreover,the fusaricidintreatmentincreasedthecatabolismoffattyandaminoacidsbutstronglyrepressedglucosedecompositionand gluconeogenesis.Insummary,ourdataprovideinsightintothemechanismoffusaricidinactivity,onwhichwebasedour suggestedstrategiesforthedevelopmentofnovelantibioticagents.
