Brzonkaliketal.AMBExpress2011,1:27 /content/1/1/27 ORIGINAL OpenAccess Processdevelopmentfortheelucidationof mycotoxinformationinAlternariaalternata KatrinBrzonkalik*,TanjaHerrling,ChristophSyldatkandAnkeNeumann Abstract TheblackmouldAlternariaalternataproducesawidediversityofmycotoxinswhichareofparticularhealth concern.SincenomaximumallowablelimitsaresetforAlternariatoxinsinfoodandfeed,preventionofAlternaria infestationsandmycotoxinspoilageistheonlywaytoavoidhealthrisks.Thus,theunderstandingofmycotoxin biosynthesisisessential.Forthatpurpose,areliablebatchprocessina2Lbioreactorwasestablishedwhich enablesthestudyofseveralparametersinfluencingtheproductionofthemycotoxinsalternariol(AOH),alternariol monomethylether(AME)andtenuazonicacid(TA)byA.alternataDSM12633.ModifiedCzapek-Doxmediumwas usedwithglucoseascarbonsourceandammoniumandnitrateasnitrogensources.Consumptionofcarbonand nitrogensourcesaswellasformationofthethreemycotoxinsweremonitored;theaveragedataoffive independentfermentationswasplottedandfittedusingalogisticequationwithfourparameters.Maximum mycotoxinconcentrationsof3.49±0.12mg/LAOH,1.62±0.14mg/LAMEand38.28±0.1mg/LTAwere obtained. Inthissystemtheeffectofdifferentaerationrates(0.53vvm-0.013vvm)wastestedwhichexertedagreat influenceonmycotoxinproduction.Theuseofthesemi-syntheticCzapek-Doxmediumallowedtheexchangeof carbonandnitrogensourcesforacetateandasparticacid.Theuseofacetateinsteadofglucoseresultedinthe soleproductionofalternariolwhereastheexchangeofammoniumandnitrateforaspartateenhancedthe productionofbothAOHandAMEwhileTAproductionwasnotaffected.
