The current study was carried out to determine the cleavage patterns of the PmpD protein in the animal pathogen C. abortus(termedPmp18D). Methodology/Principal Findings: Using antibodies directed against different regions of Pmp18D, proteomic techniques revealedthatthematureproteinwascleavedonthecellsurface,resultingina100kDaN-terminalproductanda60kDa carboxy-terminalprotein.TheN-terminalproteinwasfurtherprocessedinto84,76and73kDaproducts.Clusteringanalysis resolvedPmpDproteinsintothreedistinctcladeswithC.abortusPmp18D,beingmostsimilartothoseoriginatingfromC. psittaci,C.felisandC.caviae. Conclusions/Significance:ThisstudyindicatesthatC.abortusPmp18Disproteolyticallyprocessedatthecellsurfacesimilar totheproteinsofC.trachomatisandC.pneumoniae.However,patternsofcleavagearespecies-specific,withlowsequence conservation of PmpD across the genus. The absence of conserved domains indicates that the function of the PmpD moleculeinchlamydiaremainstobeelucidated.
