Herewereport theexperimentalcharacterization ofaproposedcausal singlenucleotidepolymorphism (SNP)in alocus relatedtoriskofCrohn’sdiseaseandulcerativecolitis.TheSNPliesintheMST1geneencodingMacrophageStimulating Protein(MSP),andresultsinanR689Caminoacidsubstitutionwithintheb-chainofMSP(MSPb).MSPbindingtotheRON receptortyrosinekinaseactivatessignalingpathwaysinvolvedintheinflammatoryresponse.Wehavepurifiedwild-type and mutant MSPb proteins and compared biochemical and biophysical properties that might impact the MSP/RON signaling pathway. Surface plasmon resonance (SPR) binding studies showed that MSPb R689C affinity to RON is approximately10-foldlowerthanthatofthewild-typeMSPbanddifferentialscanningfluorimetry(DSF)showedthatthe thermalstabilityofthemutantMSPbwasslightlylowerthanthatofwild-typeMSPb,by1.6K.Thesubstitutionwasfound not to impair the specific Arg483-Val484 peptide bond cleavage by matriptase-1, required for MSP activation, and mass spectrometryoftrypticfragmentsofthemutatedproteinshowedthatthefreethiolintroducedbytheR689Cmutationdid notformanaberrantdisulfidebond.Toge
