Twogoalswereset:1)ComparisonofACfluidfromindividualpatientstodeterminewhetherornottemporalAC isahomogenouscondition;and2)EvaluatetheproteincontentofapoolofACfluidfromseveralpatientsand qualitativelycomparethiswithpublishedproteinlistsofcerebrospinalfluid(CSF)andplasma. Methods:ACfluidfrom15patientswithtemporalACwasincludedinthisstudy.IntheACproteincomparison experiment,ACfluidfrom14patientswasdigested,analyzedbyLC-MS/MSusingasemi-quantitativelabel-free approachandthedatawerecomparedbyprincipalcomponentanalysis(PCA)togainknowledgeofprotein homogeneityofAC.IntheACproteomeevaluationexperiment,ACfluidfrom11patientswaspooled,digested,and fractionatedbySCXchromatographypriortoanalysisbyLC-MS/MS.Proteinsidentifiedwerecomparedto publisheddatabasesofproteinsidentifiedfromCSFandplasma.ACfluidproteinsnotfoundinthesetwo databaseswereexperimentallysearchedforinlumbarCSFtakenfromneurologically-normalpatients,byatargeted proteinidentificationapproachcalledMIDAS(MultipleReactionMonitoring(MRM)initiateddetectionand sequenceanalysis). Results:WedidnotidentifysystematictrendsorgroupingofdataintheACproteincomparisonexperiment, implyinglowvariabilitybetweenindividualproteomicprofilesofAC. IntheACproteomeevaluationexperiment,weidentified199proteins.Whencomparedtopreviouslypublishedlists ofproteinsidentifiedfromCSFandplasma,15oftheACproteinshadnotbeenreportedineitherofthese datasets.Byatargetedproteiniden
