However, the tissue dissemination and excretion pathway of the pathogen in goats are still poorly understood.TobetterunderstandQfeverpathogenesis,weinoculatedgroupsofpregnantgoatsviatheintranasalroute witharecentDutchoutbreakC.burnetiiisolate.Tissuedisseminationandexcretionofthepathogenwerefollowedforupto 95daysafterparturition.Goatsweresuccessfullyinfectedviatheintranasalroute.PCRandimmunohistochemistryshowed strongtropismofC.burnetiitowardstheplacentaattwotofourweeksafterinoculation.Bacterialreplicationseemedto occurpredominantlyinthetrophoblastsoftheplacentaandnotinotherorgansofgoatsandkids.TheamountofC.burnetii DNAintheorgansofgoatsandkidsincreasedtowardsparturition.Afterparturitionitdecreasedtoundetectablelevels: after81dayspost-parturitioningoatsandafter28dayspost-parturitioninkids.Infectedgoatsgavebirthtoliveordead kids.HighnumbersofC.burnetiiwereexcretedduringabortion,butalsoduringparturitionoflivebornkids.C.burnetiiwas notdetectedinfaecesorvaginalmucusbeforeparturition.Ourresultsarethefirsttodemonstratethatpregnantgoatscan beinfectedviatheintranasalroute.C.burnetiihasastrongtropismforthetrophoblastsoftheplacentaandisnotexcreted beforeparturition; pathogenexcretionoccursduringbirthofdeadaswellashealthyanimals.Besidesabortions,normal deliveriesinC.burnetii-infectedgoatsshouldbeconsideredasamajorzoonoticriskforQfeverinhum
