Results:WhileE.coliNissleisproducingAI-2inadensitydependentmanner,noAI-1wasproduced.Tostudythe effectofAI-2intheDSS(dextransulphatesodium)inducedmousemodelofacutecolitis,wesilencedthe correspondinggeneluxSbyintroninsertion.ThemutantbacteriumE.coliNissle::luxSwasequallyeffectivein colonizingthecolonandthemutationturnedouttobe100%stableduringthecourseoftheexperiment.Isolating RNAfromthecolonmucosaandperformingsemiquantitativeRTPCR,wewereabletoshowthattheexpressionof thepro-inflammatorycytokineIFN-ywassuppressedinmicebeinginfectedwiththeE.coliNisslewildtype.Mice infectedwiththeE.coliNissle::luxSmutantshowedasuppressedexpressionofIL-10comparedtouninfectedmice, whiletheexpressionofthepro-inflammatorycytokinesIL-6andTNF-αwashigherinthesemice.Theexpressionof mBD-1wassuppressedinmicebeinginfectedwiththemutantincomparisontothemicenotinfectedorinfected withthewildtype.Nodifferenceswereseeninthehistologicalexaminationofthecolonsectionsinthedifferent groupsofmice. Conclusions:E.coliNissleisproducingAI-2molecules,whichareinfluencingtheexpressionofcytokinesinthe mucosaofthecolonintheDSSmice.However,ifQShasadirectinfluenceontheprobioticpropertiesofE.coli Nissleremainstobeelucidated.
