While much is known about the regulation of Arp2/3, less is known about regulators of UNC-115/abLIM. Here we show that the Caenorhabditis elegans counterpartoftheReceptorforActivatedCKinase(RACK-1)interactsphysicallywiththeactin-bindingproteinUNC-115/ abLIMandthatRACK-1isrequiredforaxonpathfinding.GeneticinteractionsindicatethatRACK-1actscell-autonomouslyin theUNC-115/abLIMpathwayinaxonpathfindingandlamellipodiaandfilopodiaformation,downstreamoftheCED-10/Rac GTPaseandinparalleltoMIG-2/RhoG.Furthermore,weshowthatRACK-1isinvolvedinmigrationofthegonadaldistaltip cellsandthatthesignalingpathwaysinvolvedinthisprocessmightbedistinctfromthoseinvolvedinaxonpathfinding.In sum, these studies pinpointRACK-1 as acomponentof anovel signaling pathway involvingRac GTPasesand UNC-115/ abLIMandsuggestthatRACK-1mightbeinvolvedintheregulationoftheactincytoskeletonandlamellipodiaandfilopodia formationinmigratingcellsandgrowthcones.
