rBPI21 PromotesLipopolysaccharideAggregationand ExertsItsAntimicrobialEffectsby(Hemi)fusionof PG-ContainingMembranes MarcoM.Domingues,MiguelA.R.B.Castanho,NunoC.Santos* InstitutodeMedicinaMolecular,FaculdadedeMedicinadaUniversidadedeLisboa,Lisboa,Portugal Abstract Antimicrobial peptides (AMPs) are important potential alternatives to conventional therapies against bacterial infections. rBPI21isa21kDapeptidebasedontheN-terminalregionoftheneutrophilbactericidal/permeability-increasingprotein(BPI). ThisAMPpossesseshighlyselectivebactericidaleffectsonGram-negativebacteriaandhaveaffinityforlipopolysaccharide (LPS)whichisbelievedtobeattheoriginofitsneutralizingeffectoftheLPSsegregatedintothebloodstream.Weaimat understandingthemolecularbasesofrBPI21bactericidalandLPSneutralizationactions,usingbiomembranemodelsystems. UsingdynamiclightscatteringspectroscopywedemonstratethatrBPI21promotesaggregationofnegativelychargedlarge unilamellarvesicles(LUV),evenintheabsenceofLPS,andLPSaggregates,whileforzwitterionicphosphatidylcholine(POPC) LUV the size remains unchanged. The peptide also promotes the fusion (or hemifusion) of membranes containing phosphatidylglycerol(POPG).TheaggregationandfusionofnegativelychargedLUVarepeptideconcentration-dependent untilmassive aggregation is reached,followed by sampleflocculation/precipitation. Concomitantly, thereis a progressive changeinthezeta-potentialoftheLUVsystemsandLPSaggregates.LUVsystemscomposedofphosphatidylglycerol(POPG) andlipidmixtureswithPOPGhavehigherzeta-potentialvariationsthanintheabsenceofPOPG.TheinteractionofrBPI21 with lipidvesiclesisfollowedbyleakage,withhighereffectinPOPG-containingmembranes.LPSaggregationcanberelatedwitha decreasedtoxicity,possiblybyfacilitatingitsclearancebymacrophagephagocytosisand/orblockingofLPSspecificreceptor recognition.OurdataindicatethatrBPI21mechanismofactionatthemolecularlevelinvolvestheinteractionwiththeLPSof theoutermembraneofGram-negativebacteria,followedbyinternalizationandleakageinductionth
