Methods: Using small-angle X-ray scattering, measurements of collagen D-periodicity, fibril diameter and interfibrillar spacingweremadeat1mmintervalsacrosssixnormalpost-mortemcorneas(twoabovephysiologicalhydration(swollen) andfourbelow(unswollen))andtwopost-transplantkeratoconuscornealbuttons(oneswollen;oneunswollen),beforeand afterhypo-osmolarcross-linking.Thesameparametersweremeasuredinthreeotherunswollennormalcorneasbeforeand afteriso-osmolarcross-linkingandinthreepairsofswollennormalcorneas,inwhichonlytheleftwascross-linked(withiso- osmolarriboflavin). Results:Hypo-osmolarcross-linkingresultedinanincreaseincornealhydrationinallcorneas.Inthekeratoconuscorneas andunswollennormalcorneas,thiswasaccompaniedbyanincreaseincollageninterfibrillarspacing(p,0.001);anincrease in fibril diameter was also seen in two out of four unswollen normal corneas and one unswollen keratoconus cornea (p,0.001).Iso-osmolarcross-linkingresultedinadecreaseintissuehydrationintheswollennormalcorneasonly.Although therewasnoc
