RiboflavinAmelioratesCisplatinInducedToxicitiesunder Photoillumination IftekharHassan1,SandeshChibber1,AijazA.Khan2,ImranaNaseem1* 1DepartmentofBiochemistry,FacultyofLifeSciences,AligarhMuslimUniversity,Aligarh,India,2DepartmentofAnatomy,JawaharlalNehruMedicalCollege,Aligarh MuslimUniversity,Aligarh,India Abstract Background:Cisplatinisaneffectiveanticancerdrugthatelicitsmanysideeffectsmainlyduetoinductionofoxidativeand nitrosativestressesduringprolongedchemotherapy.Theseverityofthesesideeffectsconsequentlyrestrictsitsclinicaluse underlongtermtreatment.Riboflavinisanessentialvitaminusedinvariousmetabolicredoxreactionsintheformofflavin adeninedinucleotideandflavinmononucleotide. Besides,ithasexcellentphotosensitizing propertythatcanbe usedto amelioratethesetoxicitiesinmiceunderphotodynamictherapy. Methods and Findings: Riboflavin, cisplatin and their combinations were given to the separate groups of mice under photoilluminated condition under specific treatment regime. Their kidney and liver were excised for comet assay and histopathologicalstudies.Furthermore,FourierTransformInfraredSpectroscopyofriboflavin-cisplatincombinationinvitro was also conducted to investigate any possible interaction between the two compounds. Their comet assay and histopathologicalexaminationrevealedthatriboflavinincombinationwithcisplatinwasabletoprotectthetissuesfrom cisplatininducedtoxicitiesanddamages.Moreover,FourierTransformInfraredSpectroscopyanalysisofthecombination indicatedastrongmolecularinteractionamongtheirconstituentgroupsthatmaybeassignedfortheprotectiveeffectof thecombinationinthetreatedanimals. Conclusion:Inclusionofriboflavindiminishescisplatininducedtoxicitieswhichmaypossiblymakethecisplatin-riboflavin combination, an effective treatment strategy under chemoradiotherapy in pronouncing its antineoplastic activity and sensitivitytowardsthecancercellsascomparedtocisplatinalone.
