This study aimed to investigate the effect of silencing the GRP78 and clathrin-mediatedendocytosisondenguevirusentryandmultiplicationintoHepG2cells. Methodology/PrincipalFindings:HepG2cellsweretransfectedusingspecificsiRNAstosilencethecellularsurfacereceptor (GRP78)andclathrin-mediatedendocytosispathway.Geneexpressionanalysisshowedamarkeddown-regulationofthe targeted genes (87.2%, 90.3%, and 87.8% for GRP78, CLTC, and DNM2 respectively) in transfected HepG2 cells when measuredbyRT-qPCR.IntracellularandextracellularviralRNAloadswerequantifiedbyRT-qPCRtoinvestigatetheeffectof silencing the attachment receptor and clathrin-mediated endocytosis on dengue virus entry. Silenced cells showed a significantreductionofintracellular(92.4%)andextracellularviralRNAload(71.4%)comparedtonon-silencedcells.Flow cytometryanalysisshowedamarkedreductionofinfectedcells(89.7%)insilencedHepG2cellscomparedtonon-silenced cells.Furthermore,theabilitytogenerateinfectiousvirionsusingtheplaqueassaywasreduced1.07loginsilencedHepG2 cells. Conclusions/Significance:Silencingtheattachmentreceptorandclathrin-mediatedendocytosisusingsiRNAcouldinhibit denguevirusentryandmultiplicationintoHepG2cells.Thisleadstoreductionofinfectedcellsaswellastheviralload,
