DNA sequencing is a potential remedy to overcome this taxonomic impediment. Mitochondrial DNA is most commonlyused,e.g.,forspeciesidentification(‘‘DNAbarcoding’’).Here,wepresentthefirststudyinarthropodsbasedona near-completespeciessamplingofafamily-leveltaxonfromtheentireAustralianregion.Weaimedtoassesshowreliably mtDNAdatacancapturespeciesdiversitywhenmanysisterspeciespairsareincluded.Then,wecontrastedphylogenetic subsamplingwiththehithertomorecommonlyappliedgeographicalsubsampling,wheresisterspeciesarenotnecessarily captured. Methodology/PrincipalFindings:Wesequenced800bpcox1for1,439individualsincluding260Australianspecies(78% speciescoverage).Weusedclusteringwiththresholdsof1to10%andgeneralmixedYuleCoalescent(GMYC)analysisfor the estimation of species richness. The performance metrics used were taxonomic accuracy and agreement between the morphologicalandmolecularspeciesrichnessestimation.Clustering(atthe3%level)andGMYCreliablyestimatedspecies diversityforsingleormultiplegeographicregions,withanerrorforlargercladesoflowerthan10%,thusoutperforming parataxonomy.However,theratesoferrorwerehigherforsomeindividualgenera,withvaluesofupto45%whenvery recentspeciesformednonmonophyleticclusters.Taxonomicaccuracywasalwayslower,witherrorratesabove20%anda larger variation at the genus level (0 to 70%).
