Methodology/PrincipalFindings:Inthecurrentstudy,weusedamousemodelofADandgeneticallyboosteditsabilityto quenchfreeradicalsofspecificmitochondrialorigin.WefoundthatsuchmanipulationconferredtotheADmiceprotection againstvascularaswellasneuronaldeficitsthattypicallyaffectthem.Wealsofoundthatthevasculardeficitsareimproved viaantioxidantmodulationoftheendothelialnitricoxidesynthase,anenzymeprimarilyresponsiblefortheproductionof nitricoxide,whileneuronaldeficitsareimprovedviamodulationofthephosphorylationstatusoftheproteintau,whichisa neuronalcytoskeletalstabilizer. Conclusions/Significance:ThesefindingsdirectlylinkfreeradicalsofspecificmitochondrialorigintoAD-associatedvascular andneuronalpathology.
