ArabidopsisWRKY18,WRKY40,andWRKY60transcriptionfactorsinteractbothphysicallyandfunctionallyinplant defenseresponses.However,theirroleinplantabioticstressresponsehasnotbeendirectlyanalyzed. Results:WereportthatthethreeWRKYsareinvolvedinplantresponsestoabscisicacid(ABA)andabioticstress. Throughanalysisofsingle,double,andtriplemutantsandoverexpressionlinesfortheWRKYgenes,wehave shownthatWRKY18andWRKY60haveapositiveeffectonplantABAsensitivityforinhibitionofseedgermination androotgrowth.ThesametwoWRKYgenesalsoenhanceplantsensitivitytosaltandosmoticstress.WRKY40 ,on theotherhand,antagonizesWRKY18andWRKY60intheeffectonplantsensitivitytoABAandabioticstressin germinationandgrowthassays.BothWRKY18andWRKY40arerapidlyinducedbyABA,whileinductionofWRKY60 byABAisdelayed.ABA-inducibleexpressionofWRKY60isalmostcompletelyabolishedinthewrky18andwrky40 mutants.WRKY18andWRKY40recognizeaclusterofW-boxsequencesintheWRKY60promoterandactivate WRKY60expressioninprotoplasts.Thus,WRKY60mightbeadirecttargetgeneofWRKY18andWRKY40inABA signaling.Usingastabletransgenicreporter/effectorsystem,wehaveshownthatbothWRKY18andWRKY60actas weaktranscriptionalactivatorswhileWRKY40isatranscriptionalrepressorinplantcells. Conclusions:WeproposethatthethreerelatedWRKYtranscriptionfactorsformahighlyinteractingregulatory networkthatmodulatesgeneexpressioninbothplantdefenseandstressresponsesbyactingaseither transcriptionactivatororrepressor.
