Sinceanincreaseinresistancecanbe monitoredbeforethephysicalcontactbetweenscanningprobetipandsample,thistechnique isparticularlyusefultoinvestigatethetopographyofdelicatesamplessuchaslivingcells. SICMhasshownitspotentialinvariousapplicationssuchashighresolutionandlong-time imagingoflivingcellsorthedeterminationoflocalchangesincellularvolume.Furthermore, SICMhasbeencombinedwithvarioustechniquessuchas?uorescencemicroscopyorpatch clampingtoreveallocalizedinformationaboutproteinsorproteinfunctions. Thisreview detailsthevariousadvantagesandpitfallsofSICMandprovidesanoverviewoftherecent developmentsandapplicationsofSICMinbiologicalimaging.Furthermore,weshowthatin principle,acombinationofSICMandionselectivemicro-electrodesenablesonetomonitor thelocalionactivitysurroundingalivingcell.
