Methods:COMP-C3blevelsinseraweremeasuredbyusinganenzyme-linkedimmunosorbentassay(ELISA) capturingCOMPanddetectingC3b.SerumCOMPwasmeasuredbyusingELISA. Results:COMP-C3blevelsweresignificantlyelevatedinpatientswithRAaswellasinsystemiclupus erythematosus(SLE),comparedwithhealthycontrols.SLEpatientswitharthritishadsignificantlyhigherCOMP-C3b levelsthandidthosewithout.COMP-C3bwasfurthermoreelevatedinpatientswithankylosingspondylitis(AS), psoriaticarthritis(PsA),reactivearthritis,systemicsclerosis,andOA.COMP-C3bdidnotcorrelatewithCOMPinany ofthepatientgroups.COMP-C3bcorrelatedwithdiseaseactivityinRA,butnotinotherdiseases.COMP-C3blevels inRApatientsdecreasedontreatmentwithtumornecrosisfactor(TNF)-ainhibitors,whereasthelevelsincreased inpatientswithASorPsA.ThechangesofCOMP-C3bdidnotparallelthechangesofC-reactiveprotein(CRP). Conclusions:COMP-C3blevelsareelevatedinseveralrheumatologicdiseasesandcorrelatewithinflammatory measuresinRA.COMP-C3blevelsinRAdecreaseduringTNF-ainhibitiondifferentlyfromthoseofCRP,suggesting thatformationofCOMP-C3brelatestodiseasefeaturesnotreflectedbygeneralinflammationmeasures.
