We inspected the labial salivary transcriptome and proteome of Helicoverpa armigera, an important polyphagous pest species. To identify the majorityofthesalivaryproteinswehaverandomlysequenced19,389expressedsequencetags(ESTs)fromanormalized cDNAlibraryofsalivaryglands.Inparallel,anon-cytosolicenrichedproteinfractionwasobtainedfromlabialsalivaryglands and subjected to two-dimensional gel electrophoresis (2-DE) and de novo peptide sequencing. This procedure allowed comparisonofpeptidesandESTsequencesandenabledustoidentify65proteinspotsfromthesecretedlabialsaliva2DE proteome.Themassspectrometryanalysisrevealedecdysone,glucoseoxidase,fructosidase,carboxyl/cholinesteraseandan uncharacterizedproteinpreviouslydetectedinH.armigeramidgutproteome.Consistently,theircorrespondingtranscripts areamongthemostabundantinourcDNAlibrary.Wedidfindredundancyofsequenceidentificationofsaliva-secreted proteinssuggestingmultipleisoforms.Asexpected,wefoundseveralenzymesresponsiblefordigestionandplantoffense. Inaddition,weidentifiednon-digestiveproteinssuchasanargininekinaseandabundantproteinsofunknownfunction. Thisidentificationofsecretedsalivaryglandproteinsallowsamorecomprehensiveunderstandingofinsectfeedingand posesnewchallengesfortheelucidationofproteinfunction.
