GrégoryGuirimand1,VincentCourdavault1,ArnaudLanoue1,SamiraMahroug1,2,AnthonyGuihur1,NathalieBlanc1, NathalieGiglioli-Guivarc’h1,BenoitSt-Pierre1,VincentBurlat1,3,4* Abstract Background:Thefirsttwoenzymaticstepsofmonoterpeneindolealkaloid(MIA)biosyntheticpathwayare catalysedbystrictosidinesynthase(STR)thatcondensatestryptamineandsecologanintoformstrictosidineandby strictosidineb-D-glucosidase(SGD)thatsubsequentlyhydrolysestheglucosemoietyofstrictosidine.Theresulting unstableaglyconisrapidlyconvertedintoahighlyreactivedialdehyde,fromwhichmorethan2,000MIAsare derived.Manystudieswereconductedtoelucidatethebiosynthesisandregulationofpharmacologicallyvaluable MIAssuchasvinblastineandvincristineinCatharanthusroseusorajmalineinRauvolfiaserpentina.However,very fewreportsfocusedontheMIAphysiologicalfunctions. Results:Inthisstudyweshowedthatastrictosidinepoolexistedinplantaandthatthestrictosidine deglucosylationproduct(s)was(were)specificallyresponsibleforinvitroproteincross-linkingandprecipitation suggestingapotentialroleforstrictosidineactivationinplantdefence.Thespatialfeasibilityofsuchanactivation processwasevaluatedinplanta.Ontheonehand,insituhybridisationstudiesshowedthatCrSTRandCrSGDwere coexpressedintheepidermalfirstbarrierofC.roseusaerialorgans.However,acombinationofGFP-imaging, bimolecularfluorescencecomplementationandelectromobilityshift-zymogramexperimentsrevealedthatSTR frombothC.roseusandR.serpentinawerelocalisedtothevacuolewhereasSGDfrombothspecieswereshownto accumulateashighlystablesupramolecularaggregateswithinthenucleus.Deletionandfusionstudiesallowedus toidentifyandtodemonstratethefunctionalityofCrSTRandCrSGDtargetingsequences. Conclusions:AspatialmodelwasdrawntoexplaintheroleofthesubcellularsequestrationofSTRandSGDto controltheMIAmetabolicfluxundernormalphysiologicalconditions.Themodelalsoillustratestheposs
