Method:HT-29coloncancercellsweretreatedwithgradientdilutionofTXL.Themitochondrialmembrane potentialwasmeasuredbyJC-1assay.Thereleaseofcytochromecfrommitochondrialandapoptosis-related proteinsBax,Bcl-2,cleavedcaspase-3,9wereexaminedbyWesternblotanalysis.HT-29cellswereimplantedin nudemicetoexaminetheeffectsofTXLontumorgrowth. Result:TXLinhibitedHT-29xenograftedmodelandshowedastronganddose-dependentinhibitoryeffectonthe proliferationofHT-29cells.MitochondrialmembranepotentialwasreducedbyTXLattheconcentrationof0.5% above.ForWesternblotanalysis,anincreaseinBaxexpressionandadecreaseinBcl-2expressionwereobservedin TXL-treatedcells.TXLtreatmentincreasedtheproteinlevelofcleavedcasepase-3andcaspase-9,andthereleaseof cytochromecincytoplasmwasup-regulatedaswell.
