As a result, 2–3 million children and 0.5–1 million adults succumb to the consequences of this major healthcare concern. The majority of these deaths can be attributed to toxin mediated diarrhea by infectious agents, such as E. coli, V. cholerae or Rotavirus. Our understanding of the pathophysiological processes underlying these infectious diseases has notably improved over the last years. This review will focus on the cellular mechanism of action of the most common enterotoxins and the latest specific therapeutic approaches that have been developed to contain their lethal effects. Keywords: cholera; enterotoxigenic E. coli; enterotoxin; NSP4; STa; LT-I 1. Introduction Diarrhea is widely accepted as a major healthcare concern. However, only closer examination of the epidemiological data available to us fully illustrates the massive burden that acute diarrheal illness (ADI) places on our society. Children are particularly vulnerable to the lethal effects of ADI: 3.2 episodes of diarrhea occur per child/year in the age group 5 years [1]. A total of 2.5 million ADI related deaths worldwide (5 years) occur each year [1]. This means that one out of five deaths in children (5 years) is ca
