Results:TreatmentwithtrichostatinA,whichinhibitshistonedeacetylases,resultedinincreasedhistoneH4 acetylationaccompaniedbythedecondensationofinterphasechromatinandadecreaseinbothglobalH3K9 dimethylationandDNAmethylationduringmitosisinmaizeroottipcells.Theseobservationssuggestthathistone acetylationmayaffectDNAandhistonemethylationduringmitosis.Treatmentwith5-azacytidine,acytosine analogthatreducesDNAmethylation,causedchromatindecondensationandmediatedanincreaseinH4 acetylation,inadditiontoreducedDNAmethylationandH3K9dimethylationduringinterphaseandmitosis.These resultssuggestthatdecreasedDNAmethylationcausesareductioninH3K9dimethylationandanincreaseinH4 acetylation. Conclusions:Theinterchangeableeffectsof5-azacytidineandtrichostatinAonH4acetylation,DNAmethylation andH3K9dimethylationindicateamutuallyreinforcingactionbetweenhistoneacetylation,DNAmethylationand histonemethylationwithrespecttochromatinmodification.TreatmentwithtrichostatinAand5-azacytidine treatmentcausedadecreaseinthemitoticindex,suggestingthatH4deacetylationandDNAandH3K9 methylationmaycontainthenecessaryinformationfortriggeringmitosisinmaizeroottips. Background In yeast, HATs and HDACs have been found to alter Thebasicunitofchromatinineukaryotesisthenucleo- global histone acetylation levels over large regions of some, which is composed of ~146 base pairs of DNA chromatin[4].DNAitselfmaybemodifiedthroughthe wrappedaroundanoctame
