Manyenvironmentalfactorsaffectplantdevelopmentandlimitproductivityandgeographicaldistribution.The impactoftheselimitingfactorsisparticularlyrelevantformajorcrops,suchassoybean,whichhasworldwide economicimportance. Results:HereweanalyzethetranscriptionalprofileofthesoybeancysteinedesulfurasesNFS1,NFS2andISD11 genes,involvedinthebiogenesisof[Fe-S]clusters,byquantitativeRT-PCR.NFS1,ISD11andNFS2encodingtwo mitochondrialandoneplastidlocatedproteins,respectively,areduplicatedandshoweddistincttranscriptlevels consideringtissueandstressresponse.NFS1andISD11arehighlyexpressedinroots,whereasNFS2showedno differentialexpressionintissues.Cold-treatedplantsshowedadecreaseinNFS2andISD11transcriptlevelsinroots, andanincreasedexpressionofNFS1andISD11genesinleaves.Plantstreatedwithsalicylicacidexhibited increasedNFS1transcriptlevelsinrootsbutlowerlevelsinleaves.Insilicoanalysisofpromoterregionsindicated thepresenceofdifferentcis-elementsincysteinedesulfurasegenes,ingoodagreementwithdifferentialexpression ofeachlocus.Ourdataalsoshowedthatincreasingoftranscriptlevelsofmitochondrialgenes,NFS1/ISD11 ,are associatedwithhigheractivitiesofaldehydeoxidaseandxanthinedehydrogenase,twocytosolicFe-Sproteins. Conclusions:Ourresultssuggestarelationshipbetweengeneexpressionpattern,biochemicaleffects,and transcriptionfactorbindingsitesinpromoterregionsofcysteinedesulfurasegenes.Moreover,datashow proportionalitybetweenNFS1andISD11genesexpression.
