Methods:A549cellsweretreatedwithdifferentdosesofCOPandBER,combinedwith5-FU,CPT,andTAX.Cell viabilitywasmeasuredbyanXTT(2,3-bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide)assay. IntracellularROSlevelsweredeterminedbymeasuringtheoxidativeconversionofcellpermeable2′,7′- dichlorofluoresceindiacetatetofluorescentdichlorofluorescein.MDRofA549cellswasassessedbyrhodamine123 retentionassay. Results:BothCOPandBERsignificantlyinhibitedA549cellgrowthinadose-dependentmanner.Combinationsof COPorBERwithchemotherapeuticagents(5-FU,CPT,andTAX)exhibitedastrongerinhibitoryeffectonA549cell growth.Inaddition,COPandBERincreasedROSproductionandreducedMDRinA549cells. Conclusion:Aspotentialadjuvantstochemotherapyfornon–smallcelllungcancer,COPandBERincreaseROS production,reduceMDR,andenhancetheinhibitoryeffectsofchemotherapeuticagentsonA549cellgrowth.
