Methods:LGGsupplementedmicewereeithersensitizedorallywithCMPandcholeratoxinB-subunit(CTB)as adjuvant,orintraperitoneally(IP)withCMPbutwithouttheadjuvant.MicewerethenorallychallengedwithCMP andallergicresponseswereaccessedbymonitoringhypersensitivityscores,measuringthelevelsofCMP-specific immunoglobulins(IgG1,IgG2aandIgG)andtotalIgEfromsera,andcytokines(IL-4andIFN-g)fromspleenlysates. Results:SensitizationtoCMPwassuccessfulonlyinIPsensitizedmice,butnotinorallysensitizedmicewithCMP andCTB.Interestingly,LGGsupplementationappearedtohavereducedcow’smilkallergy(CMA)intheIPgroupof mice,asindicatedbyloweredallergicresponses. Conclusions:Adjuvant-freeIPsensitizationwithCMPwassuccessfulininducingCMAintheBalb/Cmicemodel. LGGsupplementationfavourablymodulatedimmunereactionsbyshiftingTh2-dominatedtrendstowardTh1- dominatedresponsesinCMPsensitizedmice.Ourresultsalsosuggestthatoralsensitizationbytheco- administrationofCMPandCTB,asadjuvant,mightnotbeappropriatetoinduceCMAinmice.
