ABSTRACT ABSTRACT The extraction, purification and analysis methods of the total flavones and phenolic constituents in Lygodium Japonicum were studied in the paper. GC fingerprint chrogram was established initially and the main volatile components were analysed by GC/MS. The main cotents and conclusions were provided as follows: (1) Extraction of the total flavones in Lygodium Japonicum . Ultrasonic Extraction was studied considering the extraction time and solvent consumption factors,. The optimum extraction technology conditions were as follows: ethanol concentration of 70%, solid/liquid ratio of l:10(m/V), extraction temperature of 70℃, extraction time of 90 min, the yield is 2.2%. (2) Analysis of the total flavones. The content of total flavones was determined by two methods of AlCl colorimetry and NaNO -Al(NO ) -NaOH colorimetry. Compared 3 2 3 3 with the results of recovery ,precision and stability experiments, the way of NaNO -Al(NO ) -NaOH colorimetry was more accurate and stable, the optimum 2 3 3 conditions was using NaNO -Al(NO ) -NaOH as color-developing agent, after 15 min 2 3 3 color time, under the wavelength of 510 nm, a standard curve was formed, namely 2 C=0.0939A,R =0.9998. The recovery for the sample was in the range 98.8% to 100.2%. This method has been applied to the determination of the total flavones with satisfactory result. (3) Separation and Purification of the total flavones. Macroporous resin and liqulid-liquid extraction were used for separation and purification of the total fl
