槲皮素联合顺铂对人卵巢癌SKOV-3细胞增殖及凋亡的影响 彭慧霞，潘艳艳，邢兰瑛，王艳华，蔡东阁，杨雪梅，李 虹（710004西安，西安交通大学医学院第二附属医院妇产科） [摘要] 目的 探讨槲皮素单独及联合顺铂对人卵巢癌SKOV-3细胞增殖与凋亡的影响，并选取对数生长期的卵巢癌SKOV-3细胞作为研究对象，实验分组：阴性对照组（生理盐水）、槲皮素低（25 μmol/L）、中（50 μmol/L）高剂量组（100 μmol/L）、顺铂组（10 μmol/L）联合组（槲皮素50 μmol/L+顺铂10 μmol/L）。SKOV-3细胞免疫细胞化学法检测槲皮素作用于SKOV-3细胞Bcl-2及P53蛋白表达水平的变化SKOV-3细胞63.49%；流式细胞仪发现G0/G1期细胞明显增多，S期细胞明显减少细胞周期阻滞于G1期SKOV-3细胞凋亡，高剂量组的凋亡率可达29.51%；槲皮素作用后，Bcl-2及P53表达明显降低槲皮素SKOV-3细胞增殖，诱导细胞凋亡降低凋亡相关蛋白P53和Bcl-2的表达水平Experimental Study of Quercetin combined with Cisplatin on Proliferation and Apoptosis Effects of Human Ovarian Cancer Lines SKOV-3 in vitro Ping Huixia, Pan Yanyan, Xing Lanying, Wang Yanhua, Cai Dongge, Yang Xuemei, Li Hong (Department of Gynaecology and Obstetrics， the Second Affilitated Hospital of Xi’an Jiaotong University，Xi’an 710004 ) Objective To study the effect of quercetin combined with cisplstin on the proliferation and apoptosis of SKOV-3 ovarian cancer cell. Methods Ovarian cancer cell SKOV-3 were divided into 6 groups at the logarithmic growth phase. Add quercetin in quercetin group to make final concentration be 25μmol/L, 50μmol/L, 100μmol/L and called low -dose group, middle-dose group, high -dose group; Add DDP in chemothapy group to make final concentration be 10μmol/L; Add quercetin(50μmol/L )and DDP (10μmol/L) in joint group ; Add NS in negative control group. Experiments were carried out after Ovarian cancer cell being treated with drugs for 24h, 48h and 72h. The cell proliferation was evaluated by MTT assay , the rate of cell apoptosis and the cell-cycle distribution was detected by flow cytometry , and the expression of p53 and bcl-2 were analyzed by Immunocytochemistry. Results Qrercetin could inhinbit the prolifertion of SKOV-3 ovarian cancer cell in a dose and time-dependent manner and the inhibitory rate in the high-dose group at 72h is 63.49%.
