高效液相色谱法测定川东獐牙菜中獐牙菜苦苷含量[摘要] 目的：建立川东獐牙菜中獐牙菜苦苷的含量测定方法。方法：采用高效液相色谱法，Crest C18（4.6 mm×250 mm，5 μm) 色谱柱，流动相：甲醇-水（25∶75），流速：1 ml/min，检测波长：254 nm，柱温：25℃。结果：獐牙菜苦苷在0.4～2.0 μg范围内线性关系良好（r＝0.999 9），平均回收率为99.78%，RSD=1.86%（n＝6）。结论：该方法简便、准确，重现性好, 可作为川东獐牙菜中獐牙菜苦苷的含量测定方法。 [关键词] 川东獐牙菜 獐牙菜苦苷 含量测定 [中图分类号] R284.1 [文献标识码] A[文章编号]1673-7210（2011）02（b）-042-02 Determination of Swertiamarin in Swertia davidi Franch. by HPLC CAI Xingdong, YANG Jianping, HUANG Chun, SHEN Li, LI Runqin, LIU Dan, ZHANG Lexing (Chongqing Three Gorges Medicine School, Wanzhou 404020，China) [Abstract] Objective: To establish a method for the determination of Swer-tiamarin in Swertia davidi Franch. by HPLC. Methods: High performance liquid chromatography was performed on C18 column. The chromatographic conditions were as follows: methyl alcohol-water (25∶75) as mobile phase，flow rate being 1.0 ml/min, the detecting wavelength at 254 nm and column temperature being 25℃. Results: Swertiamarin had good linear relation within the range from 0.4 to 2.0 μg (r=0.999 9). The average recovery was 99.78% with a RSD of 1.86% (n=6). Conclusion: This method is convenient, accurate, reproducible, and can be used for content determinaton of Swertiamarin in Swertia davidi Franch.. [Key words] Swertia davidi Franch.; Swertiamarin; Content determination 川东獐牙菜（Swertia davidi Franch.）又名水灵芝、鱼胆草、青鱼胆草、水黄连，为龙胆科獐芽菜属植物川东獐牙菜的干燥全草。《四川省中药材标准》1987年版以鱼胆草名收载，具有清热解毒、利胆健胃等功效，民间常用于各种类型肝炎及胆囊炎等的治疗。有研究表明，川东獐牙菜的苦苷类成分有明显的抗肿瘤作用[1]，其提取物制成的注射剂对革兰阳性菌具有较强的抑制作用[2]。川东獐牙菜仅在长江上游及其乌江支流一带海拔 130～170 m的河谷地带生长，三峡库区蓄水175 m后，失去自然生长条件成为濒危药用植物，有学者对其进行了生药学和引种栽培研究，现已实现人工种植[3-6]。川东獐牙菜具有广阔的开发利用前景，而其相关研究报道却不多，为进一步深入研究、开发利用川东獐牙菜，本实验采用高效液相色谱法对川东獐牙菜中的有效成分之一獐牙菜苦苷含量进行了测定，为川东獐牙菜的质量控制提供了参考。现将实验内容介绍如下： 1 仪器与试药 1.1 仪器 岛津LC-2010A高效液相色谱仪，紫外检测器，BSA124S-CW型万分之一电子天平（德国赛多利斯）；MILLI-Q BIOCEL超纯水机（法国密理博公司）；KQ-250DA型数控超声清洗仪（昆山市超声仪器有限公司）。 1.2 试药 獐牙菜苦苷对照品（批号，购于成都普思生物科技有限公司（纯度＞98%）；川东獐牙菜药材，购于万州，经重庆三峡中药研究所沈力教授鉴定为龙胆科（Gentianaceae）獐牙菜属植物川东獐牙菜（Swertia davidi
