(1.河北联合大学基础医学院 解剖2.唐山市第二医院 关节科)摘要目的 探讨在1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)制备的帕金森病PD)小鼠模型核因子NF-κB(nuclear factor kappaB)与FADD1-转化酶样抑制蛋白(FLICE-inhibitory proteins, FLIP)之间的关系,研究信号通路的作用机制。方法 将C57BL/6N小鼠随机分为模型组、干预组和对照组。观察行为学,采用免疫组织化学和免疫蛋白印迹法观察小鼠黑质酪氨酸羟化酶(TH)、NF-κB和的表达变化;给予后对上述变化的影响。结果 与对照组相比,模型组小鼠出现典型PD症状,TH阳性神经元明显丢失、蛋白水平下降,信号通路被激活,表达明显增多,和表达也明显增加;经干预处理后,上述变化均减轻。结 论信号通路参与激活,对PD模型小鼠保护作用。

Abstract: 【Objective】To investigate nuclear factor kappaB (NF-κB)and FLICE-inhibitory proteins (FLIP) in the substantia nigra (SN) of mice with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) induced Parkinson’s disease (PD), to explore the possible mechanism of NF-κB signaling pathway in PD. 【Methods】 C57BL/6N mice were randomly divided into odel groups, inhibitor groups and control groups. Their behavioral changes were observed. Immunohistochemistry and Western blot were performed to detect the expression of tyrosine hydroxylase (TH), NF-κB, FLIP and caspase-3 in the SN and their changes following treatment with parthenolide, a specific inhibitor of NF-κB.【Results】Compared with the control mice, the mice with PD presented with typical symptoms of PD. n the model group, the number of TH-positive neurons was substantially reduced, the NF-κB signaling pathway was actived, the expression of NF-κB significantly increased; and the expression of FLIP and caspase-3 significantly increased.
