核因子-кB表达对TNF-α 及IL-8影响以及对溃疡愈合作用[摘要] 目的:探讨肿瘤坏死因子-α(TNF-α)和白细胞介素-8(IL-8)对溃疡愈合的影响。方法:用免疫组化方法观察黏膜内胃泌素(Gas)和核因子-кB(NF-кB)在溃疡3、8、25 d时的表达;用酶联免疫吸附测定法(ELISA)测定TNF-α、IL-8在溃疡3、8、25 d时的含量;并测定黏膜下炎细胞数、胃液量和胃液pH在溃疡3、8、25 d时的量。结果:3 d组的胃泌素积分光密度、核因子-кB积分光密度、TNF-α含量、IL-8含量、炎细胞数和胃液量高于8 d组,8 d组高于25 d组;3 d组的胃液pH低于8 d组,8 d组低于25 d组。结论:胃酸分泌减少,导致炎症减弱,再通过某种机制引起NF-кB减少,进而使TNF-α和IL-8减少,TNF-α和IL-8减少又使炎细胞减少,导致溃疡逐渐愈合。 [关键词] 胃溃疡;核因子-кB;肿瘤坏死因子-α;白细胞介素-8 [中图分类号] R573.1 [文献标识码]A [文章编号]1674-4721(2010)05(b)-005-04 The effect of nuclear factor-κB expression on TNF-α and IL-8 and role on ulcer healing CHEN Jingxian, ZHANG Cuixiang, DENG Zhihui, SUN He, FU Xueying, PEI Xiaoqing, WANG Rui , WANG Guozhong* (Qiqihar Medical College, Heilongjiang Province,Qiqihar, 161006, China) [Abstract] Objective: To explore the effect of tumor necrosis factor-α (TNF-α) and interleukin-8(IL-8) on ulcer healing.Methods: Expressions of gastrin(Gas) and nuclear factor-кB(NF-кB) in mucosa were observed with immunohistochemistry method on 3 days, 8 days and 25 days following ulcer induction; The contents of TNF-α and IL-8 were determined with enzyme linked immunosorbent assay (ELISA) on 3 days, 8 days and 25 days following ulcer induction;The number of the inflammatory celles in submucous tissue, the volume of gastric juice and pH of gastric juice were also measured on day 3, 8 and 25 days following ulcer induction. Results: The integral light density of Gas and NF-кB,the content of TNF-α and IL-8, the number of the inflammatory celles and the volume of gastric juice on day 3 were more than those on day 8; those on day 8 were more than those on day 25; pH of gastric juice on day 3 was less than that on day 8; that on day 8 was less than that on day 25. Conclusion: Gastric acid secretion decreasing make inflammation weaken; inflammation weakening make NF-кB decrease by some mechanisms, and then TNF-α and IL-8 become decreasing, which results in inflammatory celles decreasing,and then ulcer heals.
