ABSTRACT Study for the Effects of Liuwei Dihuang Decoction on Hepatic Mitochondria Proteome of Kidney-yin Deficiency Rat Induced by Hyperthyrea Objectives:In this experiment,two-dimensional electrophoresis,spectrometry and computer assisted image analysis system will be used to separate and analyse the differences of liver mitochondrial proteome between normal rats,kidney-yin deficiency rats,and the kidney-yin deficiency rats treated with nourishing kidney-yin drugs.The changes of quantities and/or qualities of protein expression between this three groups (normal,disease,treated) will be revealed at the point of energy metabolism,which can be used to investigate the pathogenesis of kidney-yin deficiency and the biological action mechanism of Liuwei Dihuang Decoction. Methods:Eighteen male Wistar rats were randomly divided into three groups: normal group,kidney-yin deficiency group,Liuwei Dihuang Decoction treating group.Rats in normal group received 0.9% NaCl by intragastric administration for 21 days,while rats in disease group received thyroid tablet for 21 days.The treated group rats received thyroid tablet for 21 days by intragastric administration,while received nourishing kidney-yin drugs for the last 14 days.After the rats were killed,the liver mitochondrial were separated by differential centrifugation,and the mitochondrial proteins were extracted.Then three samples run in three gels in each group,and then the whole six gels were analysed with Image Master 2D Platinum 6.0 software.The selected protein spots were excised out,and identified by M
