J South Med Univ, 2014, 34(8): 1167-1170 doi 10.3969/j.issn. 1673-4254.2014.08.18 ·1167 · 基础研究 三种制作三种制作Balb/cBalb/c 小鼠下肢缺血模型方法的比较小鼠下肢缺血模型方法的比较 1 2 1 陈永林 ，张景科 ，孙少华 1 2 兰州大学 第一医院病理科，实验动物中心，甘肃 兰州 730000 摘要：目的采用高位结扎并离断一段股动脉、单纯性高位结扎和单纯性低位结扎股动脉三种术式，制作Balb/c 小鼠下肢缺血模 型。通过比较3种术式的优缺点，为制作用于再生医学研究的缺血模型提供理论依据。方法 选8周龄的雄性Balb/c 小鼠30 只， 随机分成3 组，分别按3 种方法制作缺血模型。术后连续观察5 d。第2 天行激光多普勒检测。结果 激光多普勒显示3 种模型 中手术侧的下肢血流阻滞。高位结扎并离断股动脉组中小鼠整个肢体坏疽并脱落；单纯性高位结扎组小鼠中出现小腿及脚爪 坏疽；单纯性低位结扎小鼠脚掌干燥并有脱皮现象，个别脚爪坏死，组织学见大片肌组织坏死及再生俢复反应。结论 单纯性低 位结扎股动脉是制作Balb/c 小鼠下肢缺血模型比较好的方法。 关键词：下肢缺血；结扎；Balb/c 小鼠，血管再生 Comparison of three approaches to establishing Balb/c mouse models of hind-limb ischemia 1 2 1 CHEN Yonglin , ZHANG Jingke , SUN Shaohua 1 2 Department of Pathology, First Hospital , Laboratory Animal Center , Lanzhou University, Lanzhou 730000, China Abstract: Objective To compared 3 approaches to establishing Balb/c mouse models of hind-limb ischemia suitable for studies of regenerative medicine. Methods Thirty eight-week-old male Balb/c mice were randomly divided into 3 groups for establishing models of hind-limb ischemia by both ligating and transecting the femoral artery proximal to the origin of the deep femoral artery branch, ligating the femoral artery proximal to the origin of the deep femoral artery branch, or ligating the femoral artery distal to the femoral artery trunk. The mice were continuously observed for 5 days after the operation, and hind-limb blood perfusion was monitored by laser Doppler imaging on the second postoperative day.
