AbstractObjectiveTo studythetherapeuticeffect ofAuroraBkinaseinhibitors、VX-680 and the cisplatin on the nudemicetransplants of hepaticcarcinoma,and observetheeffect to thenudemicetransplants of hepaticcarcinomaand the chemo-sensitivity.MethodsRandomlydivide 24 malenudemiceinjected with the human hepatoma cell lineHepG-2 into 4groups:combination group,VX-680group,cisplatin groupand control group.Sincethe7thdayafterexperimental tumorinoculation, GroupVX-680, with the doseof 12.5 mg/kg, intraperitoneal injection fortwo timesoneday, 14times totally; cisplatin, two times everyweek,40mg/kg, intraperitoneal injection,oneweek in a row, the 7th and the 14th dayeach time.Thecontrolgroupwas injectedwith normal saline, 40mg/kgand the duration ofaweek, then measurethe line ofapsides of thetumorgrowth in nude miceand measurethegross tumorvolume, in thenext day. Oneweekafter drug withdrawal, completelyremovethetumormass from thedead mice, studyinto the impact oftumor and thegrowthofhepatocellularcarcinoma in nudemice,bytextingAurorain combination group,VX-680, cisplatin groupandcontrol group, andBcl-2B, P53with Western blot and immunohistochemical method .ResultsVX-680 andcisplatinboth wereableto makethe tumor volume reduce significantly, significantreduction in the combination group(P0.05). Detected by Western blot and immunohistochemical method, in the combination groupAuroraBkinase expression decreased,and in the combination group,AuroraBkinaseinhibitors expression decreased, P53 expression increased, Bcl-2 expression decresed more significantly(P0.05) .Conclusion1.AuroraBkinaseinhibitorsVx-680 can significantlyinhibit the growth ofthe murine tumor cells of HEPG-2 bearingcancer. 2.When theAuroraBkinase inhibitorsVX-680 is combined with the conventional chemotherapydrugsDDP, the combination can in creasethe chemo-seasitivityof the hepatomacarcinomasignificantly. 3.When theAuroraBkinaseinhibitorsVx-680 is combined with DDP, Bcl2 expression decreased, P53 expression increased, so
