Expression andRoleofConnexin-43 in BrainTissue ofHypoxic-IschemicNeonatal RatsPostgraduateLin JieYing SupervisorProf.Lin NiYangAbstractObjectiveToinvestigatetheexpressionofconnexin-43(Cx43)in brain tissueof hypoxic-ischemicneonatalratsandexploreitsroleinhypoxic-ischemicbraindamage (HIBD).MethodsTotally128seven-day-oldneonatalratswererandomlydividedintotwogroups：controlgroupandhypoxic-ischemia(HI)groupwith64ratsineach. Accordingtothetimeofsacrefice,64ratsofeverygroupwerefurtherrandomly dividedintofourgroupsincludingsixhour(6h),twenty-fourhour(24h),forty-eighthour(48h)andseventy-twohour(72h),with16ratsineachgroup.Theleftcommon carotidarteryofratswereligatedfirstlyandthentheratswereputintothehypoxic cabinwiththemixedgasesofN2andO2for2hours.Theratsincontrolgroupwere cuttheskinofneckwithoutligatingtheleftcommoncarotidartery.The concentration ofO2whichwaskeptbetween7%-9%onaverageinhypoxiccabinwasmonitored byoxygenmeter.Ineachgroup,8ratswereusedforobservingpathologicchanges, neural cell apoptosis, andtheexpressionof Cx43throughthetechniquesofHE staining,TUNELstainingandimmunohistochemicalstaining,andthebrainsanother 8ratswasusedformeasuringtheexpressionlevelsofCx43proteinofratcortexand hippocampus byWestern-blot analysis.ResultThebehavior ofrats after hypoxic-ischemia (HI)：AllratsbecamedysphoriaabouttenminutesafterHI,andthenemergedcyanosisand dyspneaaftertenminutes.Afterthirtyminutes,theratsstoodunsteadily andappeared limpwithrightposteriorlimb,followingallrat’sactivitiesdecreasedobviouslyand fellintoadrowsiness.Mostofratsbehavedturningtotheleftstably2hourafter normal concentration ofoxygenwasgiven.Thepathological changes byHE staining:Observationbylightmicroscope:Thestructuresofnervecellsincontrolgroupwere normalandclear.InHIgroup,somenervecellsweredegenerated,necrosisand broken.Thestructureoftissuewasnotclear.Histologicalstudyshowedthatthe degeneratedandnecroticneuronsincreasedprogressivelyat6hto72hafterHI .The injuriesin HI72 hgroupweremostsevere.Apoptotic cellswereobse
