Exposuretherapy isoften beused totreat PTD, however,about40percentofPTSDpatients’extinctionmemoryreappear.Theprincipleof exposuretherapyisbasedonconditionalfearextinctiontheory.Sotheresearchfor mechanismofextinctionmemoryretentionwillbeofvitalsignificanceforpromotingtheclinicalPTSDtreatment.Hippocampusisanimportantbrainareaforlearningandmemorywithlongtermpotentialeffect(LTP)astheneuralbasisoflong-termmemory.TherearecloserelationshipbetweenPTSDandabnormalitiesofstructureandfunctioninhippocampus.BDNFcanpromotegrowthanddifferentiationofneurons,promotesynapticplasticityoflong-termmemory,andplayanimportantroleinthelongtermretentionofconditionalfearextinctionmemory.BDNFinhippocampusislikelyanimportantfactorforthelongtermretentionfor conditionalfearextinctionmemory.ThroughD-serine(DCS)systemsadministration,thisresearchobservaterelationshipsbetweenextinctionretentionscoresandchangesofTrkBandBDNFinhippocampusatdifferenttimepointsafterextinctiontraining,whichwillprovideexperimentalevidencesforfurtherunderstandingoftheextinctionmechanism,lookingfornewtargetsforthetreatmentofPTSDandimprovingclinicaltreatmenteffectforPTSD.MethodRatswererandomlydividedintoseveralgroups:normalcontrolgroup(withoutextinctiontraining),extinctioncontrolgroup,extinctiongroup,DCS+extinctiongroup (intraperitoneal DCS injection before extinction training) and extinction + DCS(intraperitoneal DCSinjectionafter extinctiontraining).
