基质细胞 Cav-1 表达影响骨髓瘤细胞对蛋白酶体抑制剂的敏感性英文摘要The Cav-1 expression of stromal cells affects the sensitivity of multiple myeloma cells to the proteasome inhibitor AbstractPart I Construction and Identification of Eukaryotic Plasmid Expression Vectors Encoding the shRNA Targeting Human Cav-1 and Its Stable Expression in HUVECObjective: To construct the eukaryotic plasmid vector mediated shRNA interference targeting Cav-1 gene, and transfect it into HUVEC,then screen stably transfected clonal cell line.Methods: Three plasmid expression vector （ pGPHI/Neo-Cav-1-homo 526 、pGPHI/Neo-Cav-1 homo 548、pGPHI/Neo-Cav-1-homo 781）coding for shRNA targeting Cav-1 and a control vector were designed. HUVECs stably expression Cav-1 shRNA and control- shRNA were screen with G418, as the experimental group and vector control group respectively. The cells were not transfected as the negative control group. Cav-1 expression was detected by using western blot and confocal microscopy.Results: HUVECs that stable expressed Cav-1-shRNA(experimental group) named as HUVECcav-1-knowndown, and that stable expressed control-shRNA (vector control group) named as HUVECcav-1-control. Cav-1 expression was notably down-regulated after transfecion of Cav-1-shRNA in HUVEC. Recombinant plasmid pGPHI/Neo-Cav-1-homo 781 had the strongest effects as in comparison with others. The relative value of Cav-1 is0.2199±0.0288. Compared with the control group, the expression of Cav-1 was decreasedsignificantly in experimental group (p0.01). There is no statistically difference between negative control and vector control group.
