ExpressionandmethylationofGPC5anditsclinicalsignificanceinlungadenocarcinomaAbstractBackgroundandobjectives:LungcancerremianstheleadingcauseofdeathformalignanttumorsinChina.Adenocarcinomaisthemostfrequenttypeoflungcancer.Theoccurrenceanddevelopmentoflungcancerisacomplexprocesswithmultiplestagesaffectedbymultiplefactorsandinvolvedchangesofmanygenes.Someoncogenesandtumorsuppressorgeneshavebeenidentified,andprogressionshavebeenmadeinpreventionandtreatmentoflungcancer.However,themajorityoflungcancerpatientsarediagnosedatadvancedstages,andtheoveralltreatmenteffectisstillpoor;thefive-yearoverallsurvivalrateisonlyabout15%.Themainreasonisthemechanismoflungcanceremainsunclear.Thus,furtherresearchonthemolecularmechanismsofoccurrenceanddevelopmentoflungcancer,andlookingfornewtargetsofdiagnosisandtreatmenthavebecometheurgentpriorityforimprovingtheeffectsofpreventionandtreatment.AgenomicwideassociationstudyfoundthatthegeneticpolymorphismofGPC5wassignificantlyassociatedwithlungadenocarcinoma.TofurtherexplorethemechanismofGPC5inlungcancer,wewilldetectandanalyzethedifferentexpressionandmethylationofGPC5betweenlungadenocarcinomaandpairednormaltissue,andexploretheregulatoryroleofthemethylationintheGPC5geneexpressionandtheirpotentialclinicalfeatures.Theresultsofourstudywillprovidetheoreticalbasistorevealthemolecularmechanismsoflungcancerandtargetedpreventionandtreatmentforlungadenocarcinoma.Methods:Weusedreal-timequantitativePCRtodetectexpressionlevelofGPC5in39lungadenocarcinomaandpairednormaltissues,andanalyzedthedifferencesofexpressionanditscorrelationwithclinicalfeatures.Wefurthervalidatedourresultsusingtwodatasets2fromGEOdatabase.BisulfitesequencingPCR(BSP)wasusedtodetectthemethylationoftumorandpairednormaltissuesfrom30lungadenocarcinomapatients.Wealsoanalyzedthedifferencesofmethylationbetweentumorandnormaltissueanditscorrelationwithclinicalfeatures.WeanalyzedtheassociationofGPC5geneexpressionandoverallsurvival(OS)oflungadenocarcinomabyusingonlinesurvivalanalysistool.Logranktestan
