ABSTRACTObjective:ToexploreHMGB1、NF-κBandInflammasomeNLRP3、ASCandAIM2expressionlevelsandtherelationshipwiththeoccurrence，developmentandprognosisinpatientswithacuteleukemia，itmaybeprovidelaboratoryevidencefordiagnosis、treatmentandrecurrencepredictionofacuteleukemia.Methods:Wecollectedthebloodandbonemarrowsamplesfrom18casesofALLand42casesofANLLatotalof60patientswithacuteleukemia(experimentalgroup)and20patientswithoutmalignancedisease(controlgroup)intheHematologyDepartmentoftheFirstAffiliatedHospitalofGannanMedicalCollegefromJuly2013toJuly2014.Theexperimentalgroupwasdividedintountreatedgroup、recrudescentgroupandcompleteremissiongroup,eachgroupwas20cases.ELISAdetectHMGB1、NLRP3、ASCandAIM2antigenlevelinplasma,RT-PCRdetectHMGB1andNF-κBgenelevelinbonemarrowmononuclearcells,WesternblotdetectHMGB1andNF-κBproteinlevelinbonemarrowmononuclearcells，IHCdetectHMGB1andNF-κBexpressionlevelinbonemarrow.Results:ELISAshowedthatHMGB1antigenlevelinuntreatedgroupandrecrudescentgroupweresignificantlyhigherthanincontrolgroup（P＜0.05）,NLRP3、ASC、AIM2antigenleveldoingtheopposite,therewasnostatisticallysignificantdifferencebetweencompleteremissiongroupandcontrolgroup（P＞0.05）;andtherewasnostatisticallysignificantdifferencebetweenALLandANLL（P＞0.05）.RT-PCRshowedthatHMGB1andNF-κBgeneexpressionweresignificantlyincreasedinuntreatedgroupandrecrudes-centgroupcomparedtocontrolgroup（P＜0.05）,butnotinthecompleteremissiongroup（P＞0.05）,therewasnostatisticallysignificantdifferencebetweenALLandANLL（P＞0.05）.WesternblotshowedthatHMGB1andNF-κBproteinexpressionlevelsweresignificantlyelevatedinuntreatedgroupandrecrudescentgroup（P＜0.05）,butnotincompleteremissiongroup（P＞0.05）,therewasnostatisticallysignificantdifferencebetweenALLandANLL（P＞）.IHCshowedthatHMGB1andNF-κBismainlyexpressedinuntreatedgroupandrecrudescentgroup.HMGB1waspositivelycorrelatedwithNF-κB,butnegativelycorrelatedwithNLRP3、ASCandAIM2.ThesurvivaltimeofHMGB1overexpressionpatientsislonglythanthatofnon-overexpressionpatients.Conclusions:TheInflammasomeNLRP3
