Immune effect of combined intervention with Anti-CD20 and Ad-IL-10 on spleen in NOD miceAbstractObjectiveTo investigate the immune effect of Anti-CD20 antibody combined with adenovirus vector mediated IL-10 on spleen in NOD mouse.Methods Twenty- four female NOD mice (3～5 weeks old) were randomly divided into four groups: A、B、C and D groups ,A and B groups were respectively caudal venouslyinjected 0.5 mg intravenous of Anti-CD20 antibody for the first time, after third days, 6 days, 9days were given 0.25 mg; group B and group C espectively Ad-mIL-10 first days, 3 days, 6 days and 9 days , D group was given 0.1ml Figure physiological saline. All mice were monitored for blood glycose and the incidence of diabetes mellitus weekly, and sacrified 12 weeks after first injection. Spleens were immediately taken out, and tested the number of CD3+ T cell and CD4+ T cell and the level of IL-10 and IL-27 by immunohistochemistry.Results The number of CD3+ T cell and CD4+ T cell and the level of IL-10 and IL-27 in spleen were enhanced in A、B and C groups(P＜0.01), Compared with A and C groups, the number of CD3+ T cell and CD4+ T cell and the level of IL-10 and IL-27 in spleenwere enhanced in group B.Conclusion The administration of Anti-CD20 antibody and adenovirus vector mediated IL-10 enhanced the number of CD3+ T cell and CD4+ T cell and the level of IL-10 andIL-27 in spleen， regulated the immune microenvironment and prevented diabetesdevelopment.Postgraduate student: RenAnxia Directed by Prof. Li TangKey Words : T1DM; IL-10; Anti-CD20 ; NOD mouse目录引言············································································································· 1第 1 章材料与方法························································································ 31.1 材料 ······································································································· 31.1.1 实验动物 ···························································································
