EffectofPEDFOntheExpressionofMCP-1、IL-6inCulturedHumanRenalMesangialCellsMastercandidate:JingwenZhangSupervisor:Prof.guoliangHuangABSTRACTObiective:Toobservetheimpactofpigmentepithelium-derivedfactor(PEDF)totheexpressionofmonocytechemoattractantProtein-1(MCP-1)andinterleukin-6(IL-6)inglomerularmesangialcells.Anddiscussthepossiblemechanism.Methods:Renalmesangialcellswereincubatedwithmediumcontainingdifferentconcentrationofglucose(10、20、30mmol/L)invitrofordifferenttimes(12h、24h、48h),detecttheexpressionofMCP-1、IL-6mRNAbyReactive-transcriptionporlymerasechainreaction(RT-PCR);mesangialcellsweretreatedwithdifferentconcentrationofPEDF(10、40、160nmol/L),ThelevelsofcytokinesincludingMCP-1、IL-6inculturedmediumweremeasuredbyenzyme-linkedimmunosorbentassay(ELISA)andthemRNAweredetectedbyRT-PCR,toobservetheimpactofPEDFtothosetwoinflammatoryfactors;todetecttheexpressionofNF-κBp65byimmunofluorescenceassay.Results:(1)Mesangialcellsincubatedwithmediumof5.6mmol/LconcentrationofglucosecanexpressabasiclevelofMCP-1、IL-6.Thosetwoinflammatorywereup-regulatedunderhighglucoseconditionwithconcentrationdependentandpeakedundertheconcentrationof20mmol/L.Whenitaddsto30mmol/L,theexpressionofMCP-1didn`tincreaseanymore,whiletheIL-6decreased(P0.05).Bothofthenwereup-regulatedwithtimedependent.(2)thehigh-glucoseinducedup-regulationofMCP-1andIL-6werelargelyblockedbyPEDFwithconcentrationdependent.Whenitaddsto160nmol/L,PEDFalmostcompletelyblockedtheup-regulationofMCP-1、IL-6inducedbyhighglucose.(3)ThesignalofNF-κBwastranslocatedfromthecytoplasmtothenucleiafterincubatedwithhighglucose,PEDFlargelyblockedthenucleartranslocationofNF-κB.Conclusions:(1)highglucoseconditionup-regulatedtheexpressionofMCP-1、IL-6mRNAinmesangialcells,maycontributetothedevelopmentandprogressionofdiabeticnephropathy.(2)PEDFsuppressedtheexpressionofMCP-1、IL-6mRNAandproteininhighglucosemedium,suggestedthatPEDFagainstdiabeticnephropathymaybepartiallythroughitsanti-inflammatoryeffect.(3)PEDFinhibitsthenucleartranslocationofNF-κBinmesangialc
