ABSTRACTObjective:Toinvestigatethefunctionsandcorrelationofsmallubiquitinrelatedmodifiers-1(SUMO-1)ofRWDcontainingsumoylationenhancer(RSUME)andhypoxia-induciblefactor-1α(HIF-1α)/vascularendothelialgrowthfactor(VEGF)withpituitaryadenoma.ToexploretherelationshipofexpressionbetweenRSUME,SUMO-1,HIF-1and/VEGFininvasivepituitaryadenomawithbiologicalbehavior,inordertoantiangiogenesis,reducetheinvasivenessofpituitaryadenomasandprovideanewtheoreticalbasis.Methods:ToinvestigatethecorrelationofexpressionbetweenRSUME,SUMO-1,HIF-1α,VEGF-Ainpituitaryadenomaandtheinvasivepituitaryadenomas.AccordingtotheHardy-WilsonandKnospstandardsandtheexamoficonographyandtheintraoperativefindings,pituitaryadenomasisdividedintoinvasivepituitaryadenomasgroupsandnon-invasivepituitaryadenomasgroups.TheexpressionofproteinandmRNAlevelsofRSUME,SUMO-1,HIF-1α,VEGF-Awasdetectedbyimmunohistochemistry,westernblot,qPCRin49casesofnon-invasivepituitaryadenoma,34casesofinvasivepituitaryadenomasand9normalpituitarycapsule,andanalysedtheexpressionindifferenttypesofpituitaryadenomas.TostudythemechanismofRSUMEandHIF-1α/VEGF-ApathwayinpituitaryadenomaAftertransfectingwithsiRNA-targetingRSUMEintomousepituitaryadenomaAtT-20cells,theexpressionlevelsofRSUME,HIF-1α,VEGF-AmRNAsandSUMO-1,HIF-1αandVEGF-Aproteinsweredetectedbyreal-timefluorescencequantitative-PCRandWesternblotting,respectively.Andthentheinvasionwasmeasuredbytranswellassay.Results:ExpressionandcorrelationofRSUME,SUMO-1,HIF-1α,VEGF-AinpituitaryadenomaTheexpressionlevelsofRSUME,HIF-1α,VEGF-AmRNAsandSUMO-1,HIF-1α,VEGF-AproteinsininvasivepituitaryadenomatissuesweresignificantlyhigherthanthoseinnoninvasivepituitaryadenomatissuesandnormalpituitarytissuesTherelationshipbetweenSUMO-1andHIF-1αininvasivepituitaryadenomatissueswaspositivecorrelation(r=0.687,p0.05),andtherelationshipbetweenHIF-1αandVEGF-Awasalsopositivecorrelation(r=0.773,p0.05).MechanismofRSUMEandHIF-1α/VEGFpathwayinpituitaryadenomaThemRNAexpressionlevelsofRSUMEwereraisedobviouslyaftercobaltchlorideaffec
